It is caused by the arthropode-borne flavivirus and transmitted by the bite of the Aedes aegypti or Aedes albopictus mosquito infected with one of the four dengue virus serotypes (DENV-1, DENV-2, DENV-3, or DENV-4). In this study, a screening exercise of various Malaysian medicinal plants showed that the extracts of Lawsonia inermis, Dryobalanops aromatica, Punica granatum, Zizyphus jujuba Lam. and Zingiber zerumbet exhibited potent inhibitory activity against NS2B-NS3 serine protease. The methanol extracts of Dryobalanops aromatica showed inhibition of 99.70 % at concentration of 200 �g/mL with IC50 value of 0.30 � 0.16 �g/mL. � 2019, Malaysian Society for Parasitology. The major carotenoids important to humans are ?-carotene, ?-carotene, lycopene, lutein, zeaxanthin, and ?-cryptoxanthin. Some of the biological functions and actions of these individual carotenoids are quite similar to each other, whereas others are specific. Besides genotype and location, other environmental effects such as temperature, light, mineral uptake, and pH have been found affect carotenoid development in plant tissues and organs. Therefore, this research investigated the effects of the season and storage periods during postharvest handling on the accumulation of carotenoid in pumpkin. This study shows that long-term storage of pumpkins resulted in the accumulation of lutein and ?-carotene with a slight decrease in zeaxanthin. The amounts of ?-carotene ranged from 174.583�2.105 mg/100g to 692.871�22.019 mg/100g, lutein from 19.841�9.693 mg/100g to 59.481�1.645 mg/100g, and zeaxanthin from not detected to 2.709�0.118 mg/100g. The pumpkins were collected three times in a year; they differed in that zeaxanthin was present only in the first season, while the amounts of ?-carotene and lutein were the highest in the second and third seasons, respectively. By identifying the key factors among the postharvest handling conditions that control specific carotenoid accumulations, a greater understanding of how to enhance the nutritional values of pumpkin and other crops will be gained. Postharvest storage conditions can markedly enhance and influence the levels of zeaxanthin, lutein, and ?-carotene in pumpkin. Publication Molecular docking study of naturally derived flavonoids with antiapoptotic BCL-2 and BCL-XL proteins toward ovarian cancer treatment(Wolters Kluwer Medknow Publications, 2020) ;Ghani M.F.A. ;Othman R.Nordin N.The naturally derived flavonoids are well known to have anticarcinogenic effects. Flavonoids could be an alternative strategy for ovarian cancer treatment, due to existing platinum-based drugs are reported to develop resistance with low survival rates. Inhibition of antiapoptotic proteins, namely B-cell lymphoma (Bcl-2) and B-cell lymphoma-extra large (Bcl-xl), is the key target to stimulate apoptosis process in cancer cells. This study aimed to determine the binding interaction of five naturally derived flavonoids (biochanin A, myricetin, apigenin, galangin, and fisetin) with potential antiapoptotic target proteins (Bcl-2 and Bcl-xl). The molecular docking study was conducted using AutoDock Vina program. The binding affinity and the presence of hydrogen bonds between the flavonoids and target proteins were predicted. Our findings showed that all the flavonoids showed better binding affinity with Bcl-xl than that of Bcl-2 proteins. The highest binding affinity was recorded in fisetin–Bcl-xl protein complex (−8.8 kcal/mol). Meanwhile, the other flavonoids docked with Bcl-xl protein showed binding affinities, ranging from –8.0 to –8.6 kcal/mol. A total of four hydrogen bonds, four hydrophobic contacts, and one electrostatic interaction were detected in the docked fisetin–Bcl-xl complex, explaining its high binding affinity with Bcl-xl. The present results indicate that all flavonoids could potentially serve as Bcl-xl protein inhibitors, which would consequently lead to apoptotic process in ovarian cancers. © 2020 Wolters Kluwer Medknow Publications. Acetogenin group has attracted worldwide attention in the past few years due their biological abilities as inhibitors for several types of tumour cells. Plagioneurin B was isolated via conventional chromatography and tested for thorough mechanistic apoptosis activity on human ovarian cancer cells (CAOV-3). Its structure was also docked at several possible targets using Autodock tools software. Our findings showed that plagioneurin B successfully inhibits the growth of CAOV-3 cells at IC50 of 0.62��M. The existence of apoptotic bodies, cell membrane blebbing and chromatin condensation indicated the hallmark of apoptosis. Increase of Annexin V-FITC bound to phosphatidylserine confirmed the apoptosis induction in the cells. The apoptosis event was triggered through the extrinsic and intrinsic pathways via activation of caspases 8 and 9, respectively. Stimulation of caspase 3 and the presence of DNA ladder suggested downstream apoptotic signalling were initiated. Further confirmation of apoptosis was conducted at the molecular levels where up-regulation in Bax, as well as down-regulation of Bcl-2, Hsp-70 and survivin were observed. Plagioneurin B was also seen to arrest CAOV-3 cells cycle at the G2/M phase. Docking simulation of plagioneurin B with CD95 demonstrated that the high binding affinity and hydrogen bonds formation may explain the capability of plagioneurin B to trigger apoptosis. Publication Quranic texts retrieval in Indri(Institute of Electrical and Electronics Engineers Inc., 2014) ;Othman R. ;Wahid F.A. ;Faculty of Science and Technology ;International Islamic University Malaysia (IIUM)Universiti Sains Islam Malaysia (USIM)This paper investigated the retrieval performance of Quranic texts translation of Pimpinan Ar-Rahman in Jawi Malay (the old version of Malay Language) and Rumi Malay (the new Roman version of Malay Language) with different version of Malay Language and letter scripting considerations. The purpose of this study is to test retrieval performance of Quranic text from various scripts of the same source. Methodology implemented was by using queries formulated from Frequently Asked Questions (FAQs) written by Muslim scholars in authenticated printed sources produced in Malaysia. 13 FAQs were selected from Utusan Melayu newspaper for Jawi Malay queries and 13 FAQs from Berita Harian newspaper for Rumi Malay queries. Keyword(s) were derived by researcher from FAQs as queries to represent each FAQs. Matching process was conducted by posting queries into Indri Retrieval. Result from matching process produced results of list of Ayat and these Ayat undergo relevance judgement process that involved three human judges to determine the relevancy of FAQ topic and Ayat, while verifying the accuracy of retrieval. Data analysis that consisted of three measures; precision, recall and overlap discovered competency of Indri for handling multi-script Quranic texts retrieval. � 2014 IEEE.
